Description

Plumbagin ( or 5-hydroxy-2-methyl-1,4-naphthoquinone) is a naphthoquinone naturally present in medicinal plants of the genus Plumbago or Diospyros . Although it has biological activities (antibacterial, antitumor), it is potentially toxic at high concentrations.

Its dosage is essential for:

  • Safer use of plant extracts containing plumbagin
  • Respect regulatory limits in cosmetics or phytotherapy
  • Compare concentration according to species, origins or extraction processes
  • Check for contamination in food supplements
  • Document natural formulations (creams, oils, tinctures)

Methodology

The analysis is carried out by high-performance liquid chromatography (HPLC) or LC-MS for accurate detection, even at trace levels. Depending on the type of matrix, several methods are possible:

  • HPLC-DAD : rapid analysis with UV detection at 254 nm
  • HPLC-MS/MS : for complex or low-content matrices
  • Extraction by organic solvents, possible purification on SPE (solid phase)

The method is validated according to ICH criteria (linearity, repeatability, specificity, LOD/LOQ).

Analyzed matrices

  • Hydroalcoholic or glycolic extracts of Plumbago zeylanica , Diospyros lotus , etc.
  • Mother tinctures or standardized extracts for phytotherapeutic use
  • Creams, essential oils, herbal cosmetic serums
  • Food supplements in liquid or solid form
  • Raw materials from potentially toxic medicinal plants

Industrial applications

  • Phytotherapy : control of active and toxic components
  • Natural cosmetics : securing formulations based on Plumbago
  • Quality control : batch evaluation or labeling verification
  • Pharmacotoxicological research : in vitro studies and regulatory dossiers
