Description
The BL-DMAC (Butanol-HCl – Dimethylaminocinnamaldehyde) method allows for the quantification of condensed tannins in plant extracts, particularly those from cranberries, grapes, cocoa, or certain barks. This colorimetric method relies on the specific reaction between the DMAC reagent and terminal flavanols (catechin-type), resulting in a color measured by spectrophotometry. It is rapid, reproducible, and suitable for crude extracts, powders, or liquid formulations. This assay is particularly useful for standardizing cranberry extracts (type A PACs) intended for nutraceutical, cosmetic, or animal nutrition applications.
The EP 6.1 method for determining PACs relies on the acid hydrolysis (Butanol-HCl) of condensed tannins, releasing anthocyanidins that are measured spectrophotometrically. This official method is used to quantify total PACs in polyphenol-rich plant extracts, particularly those derived from grape seeds, pine bark, cocoa, or cranberry. It is suitable for powders, dry extracts, liquids, and formulated products, and allows the result to be expressed as cyanidin equivalents.
