Description
Skin corrosion analysis assesses the corrosive potential of a product or substance on human skin. It relies on an internationally validated in vitro model using reconstituted human epidermal tissue. This method, governed by OECD guideline 431, provides a reliable alternative to animal testing for meeting regulatory requirements in the cosmetics, chemical, and detergent industries.
Test objective
This test determines whether a product causes irreversible skin damage, characterized by the destruction of the epidermal layers. It is used to:
- Classify corrosive substances according to the GHS or CLP classification systems
- Ensuring the safety of formulations before they are placed on the market
- Comply with regulatory requirements (REACH, cosmetics regulations, etc.)
- Avoid in vivo testing by demonstrating the absence of skin risk
OECD Methodology 431
The skin corrosion test is performed on reconstructed human skin (RhE) models. The principle involves applying the substance being tested to the tissues for a predetermined duration (3 min, then 60 min). Cell viability is then measured by spectrophotometry using a metabolic indicator (e.g., MTT).
The results allow us to classify the substance as:
- Corrosive (Category 1A, 1B, 1C)
- Non-corrosive (does not cause tissue damage)
