Description
The determination of proanthocyanidins (PACs) by the BL-DMAC (Butanol-HCl – Dimethylaminocinnamaldehyde) method allows the quantification of condensed tannins in plant extracts, particularly those from cranberry, grape, cocoa or certain barks. This colorimetric method is based on the specific reaction between the DMAC reagent and terminal flavanols (catechin type), giving a color measured by spectrophotometry. It is rapid, reproducible and suitable for crude extracts, powders or liquid formulations. This assay is particularly used for the standardization of cranberry extracts (type A PACs) intended for nutraceuticals, cosmetics or animal nutrition.
The determination of PACs according to the EP 6.1 method is based on acid hydrolysis (Butanol-HCl) of condensed tannins, releasing anthocyanidins measured by spectrophotometry. This official method is used to quantify total PACs in plant extracts rich in polyphenols, particularly from grape seeds, pine bark, cocoa or cranberry. It is suitable for powders, dry extracts, liquids or formulated products, and allows the result to be expressed in cyanidin equivalent. The determination according to EP 6.1 is particularly relevant for quality control, the standardization of natural raw materials and the validation of claimed concentrations in the nutraceutical, cosmetic and animal nutrition sectors.
